The Production of Anti-human Globulin reagent for Use in Blood Bank




Abstract
We have tried to produce anti-human globulin reagent by injection of human gam¡þma-globulin into rabbits. The human gamma-globulin was mixed with complete Fre¡þunds adjuvant for the first immunization and with incomplete Freunds adjuvant for the second and third immunization. The immunized serum was obtained by bleeding one week after the third injection and absorbed heteroagglutinins by a mixture of thoroughly washed A,B and 0 cells.
The prepared anti-human gamma-globulin serum was tested for the determination of optimal dilution and for specificity with sensitized cells.
The optimal dilution of the serum was 1:20 p and we could detect anti-C, -E,-c,-e,-s,¡þK,-k,-Fy,-Fy,-Jk,-Jk and -D with the antiserum. And we found the results of direct and indirect Coombs¢¥ tests performed with the serum were exactly same as those with the commercial antiserum.
